Orotidylate decarboxylase activity in Schizophyllum commune

Abstract

Orotidylate decarboxylase (OMP decase) activity was measured in mycelial macerates of Schizophyllum commune by a modification of the method of Beckwith et al., 1962. The uracil-requiring strains used in this study belong to two complementation groups: URA-1 and URA-2. OMP decase activities in ura-2 homokaryons and ura-2 × ura-2 uracil-requiring dikaryons were comparable to levels in wild-type homokaryons and dikaryons. No significant OMP decase activity was detected in ura-1 strains. The ura-1 strains of S. commune may be excellent candidates for transformation with a cloned OMP decase gene from yeast and/or S. commune.
